Specimen Preparation

  • Specimen must be processed within 24 hours of collection
  • Peripheral blood only, bone marrow not acceptable for sorted cells

Label all specimens with specimen type (blood or bone marrow). Bone marrow engraftment analysis by FISH, also known as XX/XY chimerism testing, involves determining the percentage of donor cells present in the recipient after bone marrow transplant (BMT) or hematopoietic stem cell transplantation (HSCT). FISH for XX/XY signals on interphase cells is done to monitor BMT/HSCT engraftment in recipients subsequent to a transplant using a sex-mismatched donor. For engraftment monitoring through molecular (DNA) genetic markers, please see test code BMPT. If testing is needed on sorted cells (CD3/CD33 or CD3/CD33/CD19/CD56), please draw 2 additional tubes of peripheral blood into sodium heparin (NaHep green-top) tube and send the completed cell sorting requisition form (OSU or Hematologics lab form) with the blood sample. Cell sorting must be done within 24 hours of blood draw. PLEASE NOTE: cell sorting can only be done on peripheral blood only; bone marrow sample cannot be used for cell sorting. Also indicate on the requisition form whether testing is needed on sorted cells ONLY or on a whole peripheral blood sample AND the sorted cells.
