Specimen Preparation
- Specimen must be processed within 24 hours of collection
- Peripheral blood only, bone marrow not acceptable for sorted cells
- Do not freeze
- Do not centrifuge
- Transport to the lab immediately after collection

Comments
Submission of a completed Bone Marrow Transplant (BMT) Genetic Test Requisition Form is required. Please call (614) 722-5321 to obtain the form. Please label all specimens with specimen type (Blood or Bone marrow) along with two patient identifiers (e.g. Name, DOB or MRN).
SORTED CELLS ORDERING:
If analysis of sorted cells is requested, please indicate on the Test Requisition Form:
- The types of cells requested: CD3/CD33 vs. CD3/CD33/CD19/CD56.
- Whether testing is requested on: 1) whole blood AND sorted cells, 2) whole blood ONLY, or 3) sorted cells ONLY.
SORTED CELLS SAMPLE COLLECTION:
- Only peripheral blood samples can be used for cell sorting. Bone marrow samples CANNOT be used for cell sorting.
- Blood samples for cell sorting must be collected in NaHep tubes. If whole blood analysis is also requested, then collect a separate EDTA tube.

Bone marrow engraftment analysis by FISH, also known as XX/XY chimerism testing, involves determining the percentage of donor cells present in the recipient after bone marrow transplant (BMT) or hematopoietic stem cell transplantation (HSCT). FISH for XX/XY signals on interphase cells is done to monitor BMT/HSCT engraftment in recipients subsequent to a transplant using a sex-mismatched donor.
